MiR-27a Functions as a Tumor Suppressor in Acute Leukemia by Regulating 14-3-3θ

چکیده
MicroRNAs (miRs) play major roles in normal hematopoietic differentiation and hematopoietic malignancies. In this work, we report that miR-27a, and its coordinately expressed cluster (miR-23a∼miR-27a∼miR-24-2), was down-regulated in acute leukemia cell lines and primary samples compared to hematopoietic stem-progenitor cells (HSPCs). Decreased miR-23a cluster expression in some acute leukemia cell lines was mediated by c-MYC. Replacement of miR-27a in acute leukemia cell lines inhibited cell growth due, at least in part, to increased cellular apoptosis. We identified a member of the anti-apoptotic 14-3-3 family of proteins, which support cell survival by interacting with and negatively regulating pro-apoptotic proteins such as Bax and Bad, as a target of miR-27a. Specifically, miR-27a regulated 14-3-3θ at both the mRNA and protein levels. These data indicate that miR-27a contributes a tumor suppressor-like activity in acute leukemia cells via regulation of apoptosis, and that miR-27a and 14-3-3θ may be potential therapeutic targets.

Changes in Expression of miR-1297 and PTEN Tumor Suppressor Gene in T-cell Acute Lymphoblastic Leukemia
Background and purpose: T-cell acute lymphoblastic leukemia (T-ALL) is a type of blood malignancy caused by changes in the precursors of T lymphocyte cells. The PTEN gene is one of the most common tumor suppressor genes that mutates in most human cancers, including T-ALL. Therefore, it is important to identify miRNAs that target the PTEN gene in T-ALL. For this purpose, in the present study, mi...

The aberrant expression of microRNAs (miRNAs) is associated with colorectal carcinogenesis, but the underlying mechanisms are not clear. This study showed that the miRNA-27a (miR-27a) was significantly reduced in colorectal cancer tissues and colorectal cancer cell lines, and that the reduced miR-27a was associated with distant metastasis and colorectal cancer clinical pathological stages-miR-2...
متن کاملMicroRNAs as a New Molecular Biomarker for Diagnosis and Prognosis of T-cell Acute Lymphoblastic Leukemia (T-ALL): A Systematic Review
MicroRNAs (miRNAs, miRs) are small endogenous non-coding RNAs that regulate the expression of protein-encoding genes at the post-transcriptional level. Several studies have described the role of miRNAs in T-cell acute lymphoblastic leukemia (T-ALL), including tumor suppressor and oncogenic miRNAs. Down-regulation of miRNA expression is a prominent feature of human malignancy. This down-regulati...
متن کاملmiR-27a and miR-27a* contribute to metastatic properties of osteosarcoma cells
Osteosarcoma (OS) is the most common primary malignant bone tumor in adolescents and young adults. The essential mechanisms underlying osteosarcomagenesis and progression continue to be obscure. MicroRNAs (miRNAs) have far-reaching effects on the cellular biology of development and cancer. We recently reported that unique miRNA signatures associate with the pathogenesis and progression of OS. O...
متن کاملOverexpression of 14-3-3θ promotes tumor metastasis and indicates poor prognosis in breast carcinoma
An isoform of the 14-3-3 protein family, 14-3-3θ has been linked with tumor cell proliferation and apoptosis. However, the role of 14-3-3θ in the progression of breast cancer remains unknown. Here, we report that 14-3-3θ plays a critical role in breast cancer metastasis and prognosis. The expression of 14-3-3θ was markedly higher in breast cancer tissues compared to adjacent normal tissues. A h...
